该酶从表达T4 DNA 连接酶基因的大肠杆菌中纯化而来，催化双链DNA或RNA的5'-磷酸末端和3'-羟基末端形成磷酸二酯键。该酶适用于平末端或粘性末端DNA的连接，修复双链DNA，RNA，DNA/RNA杂交链中的单链切口。
